Skinny Huawei smartphone shows off 6.2mm profile in Chinese certification

The FCC isn't the only agency playing with devices we don't even know exist, and its Chinese equivalent has recently had some hands-on time with an unknown Huawei smartphone, codename P6-U06. Luckily, there are a few pics and specs to accompany the filing, which tell us it weighs 120g (4.2 ounces) and measures 132.6 x 65.5 x 6.18mm (5.2 x 2.6 x 0.2 inch), meaning it could be one of the super-slim P series handsets a Huawei exec hinted at CES. We didn't see any evidence of these at MWC, but the same exec promised more was to come in 2013, possibly starting with this P6-U06.

Those dimensions house a 4.7-inch TFT screen at 720p resolution, quad-core 1.5GHz processor, 2GB RAM, an 8-megapixel camera on the back and an unusually large 5-megapixel sensor in the shooter up front. Unsurprisingly, Android 4.1.2 Jelly Bean is listed as the OS, while dual-SIM support and GSM / WCDMA radios suggest Asia as the target market (not to mention the Chinese certification). That's all we've got on the P6-U06 for now, but in lieu of official press shots, the handset strikes a couple more candid poses after the break.

Anthony Foxx in line for transportation post

WASHINGTON (AP) ? President Barack Obama on Monday will nominate Charlotte, N.C., Mayor Anthony Foxx as his new transportation secretary, a White House official said Sunday.

If confirmed by the Senate, Foxx would replace outgoing Secretary Ray LaHood.

Foxx is Obama's first black nominee among the new Cabinet members appointed for the second term. The president faced criticism early in his second term for a lack of diversity among his nominees.

The official insisted on anonymity to avoid public discussion of the pick before the official announcement.

The official noted that Foxx has led efforts to improve his city's transit infrastructure to expand economic opportunity for businesses and workers. During Foxx's term as mayor, Charlotte has broken ground on several important transportation projects, including the Charlotte Streetcar Project to bring modern electric tram service to the city as well as a third parallel runway at Charlotte/Douglas International Airport. The city has also moved to extend the LYNX light rail system to the University of North Carolina at Charlotte, the official said.

If confirmed by the Senate, Foxx would take over a department that has been at the center of Washington's debate over the impact of the so-called sequester cuts. The automatic cuts resulted in furloughs for air traffic controllers that helped cause delays at many airports.

Congress reached a deal last week to provide the Transportation Department flexibility that allowed it to end the air traffic controller furloughs.

Foxx, an attorney who has worked in several positions with the federal government, was first elected mayor in 2009. He raised his national profile last year when Charlotte played host to the Democratic Party's convention.

He also served as a member of the Charlotte City Council.

Venezuela opposition accuses government of post-vote vendetta

CARACAS (Reuters) - Venezuela's opposition is accusing state institutions of intimidating and threatening to fire employees critical of President Nicolas Maduro after a disputed election that stirred protests and a wave of violence.

Activists are outraged at two recordings that appear to show officials vowing to sack workers who support opposition leader Henrique Capriles, who narrowly lost a snap election to replace the late socialist president Hugo Chavez.

One opposition activist told Reuters she had received more than 2,000 complaints of workplace harassment, of which some 300 people said they had been fired for their political views.

Maduro allies deny the accusations, insisting Venezuelans should focus instead on charges that Capriles stirred up violent protests last week that killed eight people.

For many Venezuelans, it is a reminder of a list once used by Chavez's government to target opposition supporters.

"I will not accept members of fascist parties," Housing Minister Ricardo Molina said in a video of what appeared to be a meeting of ministry personnel.

"Whoever wants to be a member of (opposition party) Voluntad Popular ... can resign, because if they don't resign I'm going to fire them myself."

A ministry official said Molina had not responded and that for now his office had no plans to comment on the video.

Maduro kept Molina in his post when he unveiled a new cabinet on Sunday, a day after the video began circulating online.

The wave of discrimination charges came on the heels of violent opposition demonstrations demanding a full recount of the April 14 vote that Maduro won by less than 2 percentage points.

While officials say Capriles is responsible for the deaths, his camp believe the government has exaggerated the violence and included victims of common crime.

'WE'RE GOING TO FIRE YOU'

Opposition activist Delsa Solorzano says government agencies, public schools and state-run companies are using pictures on social networks or even stray comments critical of the government to target workers suspected of opposing Maduro.

She says she has received a flood of emails and calls from public employees, some of whom say they have been interrogated about their political views.

"They don't know who you voted for, but they suspect you might be an opposition sympathizer if once you posted some comment like 'Crime is really bad,'" said Solorzano, looking through an email inbox bulging with complaints.

"There are people in ministries whose job is to review Facebook and Twitter accounts."

In another video that also circulated widely online, an official in the western state of Zulia was apparently recorded telling employees in his office that campaign canvassing phone calls had revealed which of them supported Capriles.

"I'm telling you, we're going to fire you," Leonet Cabezas, a Zulia sports official, said in the audio recording. "We're not idiots ... We've identified you and we're going to get rid of you."

Cabezas later told a local newspaper his office was not going to sack anyone, and accused the legislator who released the recording of seeking to create "disturbances."

One anonymous Facebook page entitled "Denounce Traitors to the Fatherland" shows pictures of opposition marches with arrows and tags identifying the names and workplaces of public employees in the Andean state of Tachira.

"Now you see why we have to have to clean out so many state institutions!" one comment read.

'NO HARASSMENT'

Government allies say the charges are ridiculous because Venezuela's ballot is secret, meaning officials would have no way of knowing who voted for Capriles. Nearly all the complaints are anonymous, meaning they are impossible to verify.

"There is no harassment of workers," said ruling Socialist Party official Blanca Eekhout, according to local media.

"Why don't you ask about the deaths from violence spurred by Capriles, who should be accused of crimes against humanity?"

The situation is not without precedent in Venezuela. Ten years ago, Chavez allies collected a list of people who signed petitions calling for a recall referendum against him.

The opposition says that became a blacklist used to block people who later sought jobs or government contracts.

At the time, the practice was openly acknowledged by some rank-and-file "Chavistas," who described it as revenge for a botched 2002 coup that briefly ousted Chavez.

In 2005, the president himself lamented that government officials were using the list to decide "if someone gets a job or doesn't get a job" and ordered them to "bury" it.

Solorzano said the names of victims of discrimination after the latest election are being kept confidential, a few have gone on record with cases in previous months.

Evelyn Giambalvo, 40, who is on medical leave from the state-run Venezuelan Agricultural Bank, said her salary was cut off last September, the day after she posted a picture on social networks of herself with Capriles.

"The management said 'how can you publish that picture when you work at this institution?'" she said.

Her husband Freddys Diaz, 55, said he was fired from the same institution in February without explanation.

The Agriculture Ministry, which oversees the bank, did not respond to calls seeking comment.

"We're supposed to be living in a democratic country where you don't have to agree with what the government says, or much less applaud things you don't agree with," said Giambalvo.

Want a glimpse of what LG may have in store for the US market come the first of May? Chances are this latest render (pictured above) from @evleaks could very turn out to be AT&T's Optimus G Pro variant. From the looks of things, not much has changed design-wise since we reviewed the 5.5-inch global model, with the removable, high-gloss backplate still sporting that sub-surface pattern first introduced on the Optimus G. Even the arrangement of the rear camera, flash and speaker appear to have remained the same -- the only noticeable difference is the AT&T globe situated none too subtly below. Whether or not LG's reworked its internals remains to be seen, but with the event now less than a week away, we'll have all the answers soon enough.

Google buys Wavii for $30 million, mirroring Yahoo's deal

SAN FRANCISCO (Reuters) - Google Inc has acquired Wavii, the Seattle-based startup behind a news summarization app, for roughly $30 million in cash, a person with knowledge of the matter said Tuesday.

Google's successful bid came after Apple Inc had expressed interest in buying Wavii to incorporate the startup's natural language technology into Siri, Apple's voice-activated personal assistant feature, said the person, who declined to be named because the deal has not been publicly announced.

Google and Wavii declined to comment.

Google's purchase comes several weeks after Yahoo Inc paid a similar amount to acquire Summly, the news reader and Wavii competitor founded by 17-year-old Nick D'Aloisio in London.

The deals have taken out of play two small companies that sought to enhance how consumer experience news - a significant concern for Google and Yahoo, which both maintain highly trafficked news sites.

In separate interviews last year, Wavii founder Adrian Aoun and D'Aloisio acknowledged the competition between the two startups.

D'Aloisio touted Summly's superior user interface, which condenses articles into several easy-to-read paragraphs. Aoun played up his app's technology, including a proprietary algorithm that boiled down complex news stories into sentences of just a few words.

Wavii's investors included Paypal co-founder Max Levchin, former Facebook executive Dave Morin, and Fritz Lanman, a former dealmaker at Microsoft Corp.

Most of the startup's employees are expected to relocate to Google's headquarters in Mountain View, California.

News of the acquisition was first reported by Techcrunch.

A total of 90 species are recognized from the Indo-Malayan and Palearctic realms of Asia, 71 of which are described here as new species. A total of 438 photographs are included to aid in specimen identification, all exported to and available for the public from EOL. Newly discovered species are described from a range of countries including Brunei, China, Christmas Island, India, Indonesia, Japan, Laos, Malaysia, Nepal, The Philippines, Singapore, South Korea, Sri Lanka, Taiwan, Thailand, and Vietnam, while previous knowledge of the genus was confined only to The Philippines, Sri Lanka, and Australia. The study was published in the open access journal Zookeys.

Oxyscelio was first recognized as a distinct taxon in 1907, from a specimen collected in Java, Indonesia. The genus belongs to a wasp family that is represented mainly by parasitoid species. These are organisms that exhibit in essence parasitic behaviour, but unlike normal parasites they go further in sterilizing or killing the host, and sometimes even consuming it. The wasps from the Oxyscelio group presumably parasitise on the eggs of another insect. Previous researchers Jean Jacqus Kieffer and Alan P. Dodd continued to describe new species until 1931. Oxyscelio received almost no attention from that time until 1976, when Lubomr Masner, one of the authors of this study, published revisionary notes for genera of the then-recognized family Scelionidae. This resulted in a total of 19 species known from Asia.

Extensive specimen collecting in the following decades revealed that the actual number of species of Oxyscelio was much greater than previously recognized. The U.S. National Science Foundation (NSF) initiated Planetary Biodiversity Inventories (PBIs) to facilitate description of species that were known to a handful of scientific specialists but not yet officially published. Part of that initiative enabled the current study, which resulted from examination of thousands of preserved Oxyscelio specimens that had been housed in natural history collections around the world. This resulted in a more than fourfold increase in the number of species of Oxyscelio that are officially described from Asia.

Previously described species were examined to ensure that newly described species were distinct from them. This required the rediscovery of several species that had not been seen in a century or more, including the first described species of Oxyscelio, which had not been seen since its discovery in 1907. New technology enabled better examination of the morphology of these species, which had not even been photographed until the publication of this study.

40 percent of parents give young kids cough/cold medicine that they shouldn't

Apr. 22, 2013 ? Children can get five to 10 colds each year, so it's not surprising that adults often turn to over-the-counter cough and cold medicines to relieve their little ones' symptoms. But a new University of Michigan poll shows that many are giving young kids medicines that they should not use.

More than 40 percent of parents reported giving their children under age 4 cough medicine or multi-symptom cough and cold medicine, according to the latest University of Michigan Mott Children's Hospital National Poll on Children's Health. Twenty-five percent gave those children decongestants.

In 2008, the federal Food and Drug Administration issued an advisory that these over-the-counter medicines not be used in infants and children under age 2. They have not been proven effective for young children and may cause serious side effects, says Matthew M. Davis, M.D., M.A.P.P., director of the C.S. Mott Children's Hospital National Poll on Children's Health.

In response to the FDA, manufacturers of over-the-counter cough and cold products changed their labels back in 2008, to state that the medicines should not be used for children under 4 years old.

"These products don't reduce the time the infection will lasts and misuse could lead to serious harm," says Davis. "What can be confusing, however, is that often these products are labeled prominently as 'children's' medications. The details are often on the back of the box, in small print. That's where parents and caregivers can find instructions that they should not be used in children under 4 years old," Davis says

The side effects from use of cough and cold medicines in young children may include allergic reactions, increased or uneven heart rate, drowsiness or sleeplessness, slow and shallow breathing, confusion or hallucinations, convulsions, nausea and constipation.

The poll found that use of the cough and cold medicines in children age four and under did not differ by parent gender, race/ethnicity or by household income.

"Products like these may work for adults, and parents think it could help their children as well. But what's good for adults is not always good for children," says Davis.

Davis says parents need to be vigilant about reading the directions and should always call their pediatrician or health care provider about questions regarding over-the-counter medications.

"Because young children often suffer from cold-like symptoms, more research is needed to test the safety and efficacy of these cough and cold medicines in our littlest patients," Davis says.

As George W. Bush library opens, a rare meeting of presidents and rivals

WASHINGTON (Reuters) - Their rivalries have helped to define American politics for more than a quarter-century.

And sometimes the complex relationships among the only five people alive who know what it's like to be president of the United States have seemed to be straight out of a soap opera. They have called each other names and blamed one another for the nation's problems.

But when they have a rare meeting in Dallas on Thursday for the opening of former president George W. Bush's library and museum, there will be smiles for the cameras and friendly chatter by President Barack Obama and former presidents Bush, Bill Clinton, George H.W. Bush and Jimmy Carter.

It will be the first time they have all been together since January 2009, when they met in Washington a few days before George W. Bush left office and Obama was sworn in as president.

The library's dedication at Southern Methodist University will be something of a re-emergence for George W. Bush, who has preferred to stay out of the spotlight since leaving Washington after eight tumultuous years in office that followed six years as Texas governor.

"Fourteen years was enough for me," Bush told People Magazine last week. "But I do want to stay engaged in issues that matter to me."

Having served in a stressful job that always seems to age its occupants, the five men share a "common bond that supersedes any short-term policy or political differences," said Karen Hughes, a former top aide to George W. Bush.

That said, this isn't necessarily the friendliest of fraternities.

Obama, a 51-year-old Democrat, twice won election by denouncing Republican George W. Bush's handling of the presidency, from the struggling U.S. economy to wars in Iraq and Afghanistan.

Obama, the nation's first African-American president, also accused fellow Democrat Clinton of injecting race into the 2008 campaign, when Clinton was campaigning in South Carolina for his wife, Hillary, in the Democratic presidential primary.

Republican George W. Bush, 66, criticized Clinton's handling of the economy in defeating Clinton's vice president, Al Gore, in 2000. Clinton used the same economic argument in 1992 to deny a second term to Bush's father, George H.W. Bush.

It was George H.W. Bush, now 88, who had perhaps the most colorful putdown of Clinton and Gore in the 1992 presidential campaign when he said, "My dog Millie knows more about foreign policy than those two Bozos."

'YOU CANNOT GET MAD AT THE GUY'

It took a while for the edge to wear off from that campaign.

When then-President Clinton attended George H.W. Bush's presidential library opening in College Station, Texas in 1997, Clinton aides recall that Bush and his wife, Barbara, were particularly gracious.

On the other hand, their sons, George W. and Jeb, were frosty to the Clinton side, former Clinton press secretary Mike McCurry said.

Five years after Clinton had denied the elder Bush a second term in the White House by casting the Republican as out of touch on the economy, "things were still a little bit raw," McCurry said.

Now, Clinton and George H.W. Bush probably have the closest friendship within the group of presidents. They have worked together to raise money to help those stricken by a 2005 tsunami in the Indian Ocean that killed more than 230,000 people in more than a dozen countries.

"You cannot get mad at the guy," George H.W. Bush wrote of Clinton after traveling with him in 2005. "I admit to wondering why he can't stay on time, but when I see him interacting with folks my wonder turns to understanding, with a dollop of angst thrown in."

The younger Bush now gets along with Clinton, too. They worked together on Haiti earthquake relief in 2010.

"I like him, and I love his father," Clinton said during an appearance with George W. Bush in Salt Lake City last August.

The relationship between the last two presidents, Obama and George W. Bush, remains something of a work in progress, aides said.

The two rarely speak, although Obama was gracious to his predecessor last year when Bush visited the White House for the formal unveiling of his presidential portrait.

"We may have our differences politically, but the presidency transcends those differences," Obama said at the ceremony. "We all love this country. We all want America to succeed."

A senior administration official said that Obama, now in his second term, has not changed his views about what he saw as "poor policy decisions" by Bush on the Iraq war and the U.S. economy. But, the official said, Obama has an appreciation "for the enormity of the decisions that a president has to make and the burden that a president has to bear, especially when Americans lose their lives."

The Bush side, which was not happy at how Obama bashed him during the 2008 campaign, nevertheless is pleased that Obama is joining the group in Dallas.

If there is a wild card in the group, it is Carter.

Aides to Obama and those close to both Bushes and Clinton say the four of them are all a bit baffled and bemused by Carter, 88, who was president from 1977 to 1981, before the late Ronald Reagan's conservative revolution stormed Washington.

Carter has consistently criticized all of his successors, even when they have been fellow Democrats.

"The odd man out is Carter," said Ron Kaufman, a former adviser to the elder Bush.

So what do the presidents talk about when they get together? If the past is any guide, they will keep it light.

Dana Perino, who was a press secretary to George W. Bush, said after the presidents were together in 2009 she asked her boss what they had discussed.

Bush's response: "We mostly talked about our families."

5 Tips to Live Well with Epilepsy - Royal Oak, MI Patch

For people with?epilepsy, the onset of recurring seizures can be frightening. While many people with the condition live full and active lives, including driving a vehicle, working outside the home and playing sports, it may be necessary to make some lifestyle changes.

?The goal is to make sure the patient is seizure free with minimal side effects,? says?Shaila Gowda, M.D., director, Epilepsy Program, Beaumont Hospital, Royal Oak. ?Avoiding seizure triggers such as lack of sleep, alcohol and stress will help.?

In addition to following a supervised treatment plan with medication, it?s just as important to establish good habits to manage or avoid seizures. Once triggers have been identified, exposure can be limited.

According to the?Epilepsy Foundation, epilepsy and seizures affect more than 2 million Americans of all ages. Nearly 200,000 new cases occur each year.

Epilepsy is a brain disorder. When clusters of brain nerve cells, or neurons, signal abnormally they can cause unusual sensations, emotions and behavior; or convulsions, muscle spasms, seizures and loss of consciousness. Epilepsy can have many causes including illness, brain damage and abnormal brain development.

Dr. Gowda provides 5 helpful tips to follow when living with epilepsy:

Keep a healthy diet
A balanced diet should consist of a wide variety of carbohydrates, fats and proteins. Yet, there are some foods and beverages that should be avoided as they may trigger or worsen seizures. Foods with monosodium glutamate, the food additive commonly known as MSG, is an ?excitatory neurotransmitter? meaning it is a brain stimulant which can increase the severity of the seizure. Other foods and beverages with stimulants, like caffeine, should also be avoided. Dr. Gowda explains that an occasional drink will not cause a seizure, but prolonged alcohol consumption should be avoided. ?Studies suggest that alcohol withdrawal seizures most often occur seven hours after heavy drinking has stopped,? explains Dr. Gowda.

Get to bed
Poor sleep habits can lead to increased frequency of seizures. It?s important to get seven to nine hours of restful sleep each night. But, getting too much sleep is not good either. If you are feeling drowsy and not feeling energized during the day, make sure you?re not experiencing side effects of the medication. Discuss the dosage with your physician. ?Sometimes, if not diagnosed properly, sleep issues get dismissed as part of epilepsy,? says Dr. Gowda.

Exercise regularly
Those with epilepsy can experience fatigue and symptoms of depression. Getting regular, moderate amounts of exercise will improve overall physical health and fitness. An added benefit of exercise is that it indirectly boosts self-esteem ? keeping mood disorders and depression at bay.

Manage your stress
While it?s important to learn how to handle stress on a regular basis, excessive stress in never a good thing. Undue stress increases hormones which have been shown to cause seizures. ?To avoid getting overwhelmed, create an agenda of daily activities and prioritize tasks. It is important to keep some time aside for relaxation at the end of each day,? advises Dr. Gowda.

Choose the right doctor
Find a doctor who specializes in epilepsy. Those with epilepsy should be able to discuss any concerns freely, and work together with an expert to find a treatment program that works for them. People with epilepsy who understand their condition will be able to make informed decisions about lifestyle changes that are needed. At the OpenStack Summit last week, Tuesday’s keynote opened with?Dope’n'Stack E.N.T.E.R.P.R.I.S.E, a video that symbolizes the arrival of a new force of disruptors who see riches in building software and systems that will displace the legacy systems of old. Kerry pushes Turkey-Israel rapprochement

ISTANBUL (AP) ? Wrapping up a 24-hour visit to Istanbul, U.S. Secretary of State John Kerry on Sunday sought to cement and speed up an improvement in relations between Turkey and Israel as well as explore new ways to relaunch Mideast peace efforts.

President Barack Obama has made both issues foreign policy priorities for his second term and Kerry was pushing them in meetings with Palestinian leader Mahmoud Abbas and Turkish Foreign Minister Ahmet Davutoglu.

On a trip to Israel last month, Obama secured a pledge from Turkish and Israeli leaders to normalize ties that broke down after a 2010 Israeli raid on a Gaza-bound aid flotilla that killed eight Turks and a Turkish-American. However, the rapprochement has been slow, sparking concerns that Turkey may be backsliding on its commitment.

U.S. officials say they are hopeful that a meeting this week of Israeli and Turkish diplomats to discuss Israeli compensation for victims of the flotilla raid will jumpstart the process of restoring full diplomatic relations and exchanging ambassadors between the two countries that Washington sees as key strategic partners in the volatile Middle East.

Turkey's Prime Minister Recep Tayyip Erdogan is due to visit Obama at the White House on May 16 and U.S. officials are keen to see substantive process by then.

However, Erdogan's plans to visit the Hamas-controlled Gaza Strip sometime in May after his trip to Washington have raised concerns. The State Department has said such a journey would be unhelpful. Both Israel and Abbas, whose Palestinian Authority is based in the West Bank, are opposed.

With Abbas, Kerry was talking about ways to improve the Palestinians' living conditions as a confidence-building measure to improve the atmosphere for a resumption in stalled peace talks with Israel.

Kerry has said he fears there is only a two- or three-year window of opportunity to reach a deal on a two-state solution that would end the decades-old Israeli-Palestinian conflict and wants to move as quickly as possible. He has met with Abbas and Israeli Prime Minister Benjamin Netanyahu several times already to that end since becoming secretary of state.

Kerry was in Istanbul primarily to attend an international conference on Syria that began on Saturday afternoon and stretched into early Sunday morning as participants debated how best to boost aid to rebels trying to topple Syrian President Bashar Assad's regime.

At the conference, he announced that the Obama administration would double its non-lethal assistance to the Syrian opposition with an additional $123 million in supplies that could include for the first time armored vehicles, body armor, night vision goggles and other defensive military supplies.

The additional aid, which brings total non-lethal U.S. assistance to the opposition to $250 million since the fighting began more than two years ago, "underscores the United States' firm support for a political solution to the crisis in Syria and for the opposition's advancement of an inclusive, tolerant vision for a post-Assad Syria," Kerry said.

The U.S. pledge was the only tangible, public offer of new international support at the meeting of the foreign ministers of the 11 main countries supporting the opposition and fell well short of what the opposition has been appealing for: weapons and direct military intervention to stop the violence that has killed more than 70,000 people.

The Syrian National Coalition is seeking drone strikes on sites from which the regime has fired missiles, the imposition of no-fly zones and protected humanitarian corridors to ensure the safety of civilians.

While pleased with the U.S. moves, the opposition appeared deeply disappointed, especially as it lost some ground in the latest clashes with Syrian troops backed by pro-government gunmen capturing at least one village in a strategic area near the Lebanese border.

"We appreciate the limited support given by the international community, but it is not sufficient," it said in a statement released at the end of the conference. "We call on the international community to be more forthcoming and unreserved to fulfill its responsibilities in extending support that is needed by the Syrian people."

With the exception of the United States, none of the participants offered new assistance, although European nations are considering changes to an arms embargo that would allow weapons transfers to the Syrian opposition. But European Union action is unlikely before the current embargo is set to expire in late May.

Obama has said he has no plans to send weapons or give lethal aid to the rebels, despite pressure from Congress, some administration advisers and the appeals from opposition.

There are no plans to change that policy, although U.S. officials say they are not opposed to other countries sending arms, as long as the recipients have been properly vetted.

Fires on all cylinders when it drops all pretense and allows its talented cast to simply belt out a series of pure, unfiltered slices of ear candy.

A rousing soundtrack helps to compensate for some of the historical embellishments in this Australian crowd-pleaser.

New potential target for cancer therapy identified

Apr. 19, 2013 ? Researchers at UT Southwestern Medical Center have found that alternative splicing -- a process that allows a single gene to code for multiple proteins -- appears to be a new potential target for anti-telomerase cancer therapy.

The enzyme telomerase is overexpressed in almost all cancer cells, and previous research efforts have failed to identify good telomerase inhibitors. The study by Dr. Woodring Wright and UT Southwestern colleagues in the April 4 issue of Cell Reports identifies a new approach for inhibiting telomerase, which is an enzyme that drives uncontrolled division and replication of cancer cells.

Alternative splicing allows genetic information to be assembled in different ways and is almost always controlled by DNA sequences that are immediately adjacent to the parts of a gene that code for protein. "In the case of the telomerase gene, we found that these controlling regions are located very far from the protein coding regions and that they contain unusual DNA sequences," said Dr. Wright, professor of cell biology and internal medicine. "Their unusual DNA structure suggests that humans regulate telomerase in a very different fashion that we may be able to exploit to develop inhibitors of the enzyme."

Most of the splice variants that telomerase makes are inactive, but Dr. Wright's team demonstrated that it was possible to shift the splicing to make even less active telomerase, potentially providing a new approach for cancer therapy.

Telomeres are specialized structures at the ends of each chromosome. As DNA replicates, telomeres shorten each time a cell divides. Telomerase in human cancer cells is 10 to 20 times more active than in normal body cells. The increase provides a selective growth advantage to many types of tumors. If telomerase activity was to be turned off, then telomeres in cancer cells would shorten like they do in normal body cells.

"The oft-used analogy is that telomeres are like the plastic ends of shoelaces that protect them from fraying," Dr. Wright said. "Once the plastic becomes damaged and falls off, the shoelace can no longer be threaded effectively. The only solution at that point is to throw the shoelace away."

In most tissues, telomerase turns off during development, after which telomeres shorten and limit the number of times a cell can divide, eventually losing their capping function similar to the shoelace tip falling off. This timing also functions as a tumor-protection mechanism, since the limited cellular lifespan prevents pre-malignant cells from accumulating the mutations they need to become cancerous.

Preclinical studies have shown that inhibiting telomerase causes tumor cells to lose immortality, re-initiate telomere shortening, and eventually die. In the event that a tumor has already thoroughly developed, it may be surgically removed, and following conventional chemotherapy, telomerase inhibition could prevent rare surviving cells from having enough divisions to cause a relapse.

Dr. Wright said the alternative splicing method also could be useful for regenerative medicine, because telomeres in our stem cells shorten with age and that eventually compromises their function. "Under the right circumstances, increasing or decreasing telomerase activity could profoundly affect our treatments for both cancer and aging," he said.
